Prepare in screw-cap bottles as in the case of medium 667. After combining and mixing all solutions (as in the case of medium 667) adjust the pH of this medium with sterile HCl or Na2CO3 solution (2 mol/liter each) to pH 6.8. All other steps are the same as used in medium 667. Incubate in the light using a tungsten lamp. Feed periodically with neutralized solution of sodium sulfide (see medium 232) and other supplement solutions to replenish photosynthetic electron donors (see Ref. [1579]). |
